If you’re one of those researchers who has been avoiding ChIP experiments like the plague because of the aggravating low yields and poor quality ChIP DNA associated with traditional ChIP protocols, then it’s time to give the ChIP DNA Clean & Concentrator™ (ChIP DCC™) Kit from Zymo Research a whirl.
The ChIP DCC™ is a quick and effective way to purify and concentrate high quality DNA after any step in a standard ChIP protocol, including:
- Reverse cross-linking
- Proteinase K or RNase A digestion
- Mechanical or nuclease-mediated DNA shearing
- Elution from chromatin-antibody-bead complexes
Farewell to Phenol-chloroform
ChIP DCC™ technology actually does such a good job, that those hazardous (not to mention smelly) phenol-chloroform extractions may soon seem as outdated as a brick-sized cell phone. ChIP DCC™ is about more than just convenience; it helps generate more accurate and better quality data from each experiment, as revealed below in a head to head comparison conducted by Zymo Research.
ChIP DNA Purification Methods Comaprison- ChIP assays were performed with HeLa cells and ChIP-grade anti-H3K4me3 and rabbit IgG. Both total and immunoprecipitated choromatin were reverse cross-linked and recovered using either ChIP DNA Clean and Concentrator™, phenol-chloroform extraction, or DNA recover kit from supplier Q. The amount of ChIP DNA was determined using qPCR with primers specific to a region in the GAPDH promoter. ChIP DNA enrichment is graphed as % of input (i.e., the relative amount of immunoprecipitated DNA compared to input DNA after qPCR analysis).
That’s what Zymo calls Purity By Design, and the bottom line is: high quality ChIP-DNA results in high quality data.
Beta-Test the new Zymo-Spin™ ChIP Kit
The Zymo R&D team has paired up a top-of-the-line, Protein A bead based chromatin immunoprecipitation protocol with their unique micro-elution Zymo-Spin™ columns from the ChIP DCC™ to create the new Zymo-Spin™ ChIP Kit. A package that provides several benefits for ChIP, ChIP-qPCR and ChIP-seq library prep users, including:
- Fast and streamlined workflow that can be completed in 8 hrs.
- Micro-elution spin columns for enhanced ChIP DNA clean-up and recovery.
- High quality ChIP DNA that provides better results for downstream applications such as; end point PCR, quantitative real-time PCR (ChIP-qPCR), ChIP-seq library preparation, and Next-Generation sequencing.
If you’d like to be the one of the first to get your hands on the Zymo-Spin™ ChIP Kit, Zymo Research is offering early access to beta-testers if you sign up before September 18th, 2013. Qualified users will supply important pre-launch feedback while they wrap up all of the finishing touches. To apply and get more information visit the Zymo Research Zymo-Spin™ ChIP Kit page here.
